The Succinate Glo JmjC Demethylase Hydroxylase Assay is a homogeneous, double-reagent-addition method to rapidly detect succinate formation in JmjC histone demethylase and Fe(II)/alpha-ketoglutarate-dependent dioxygenase reactions. After the demethylase or dioxygenase reaction, an equal volume of Succinate Detection Reagent I and Succinate Detection Reagent II are added sequentially to convert the succinate product to ATP and generate light in a luciferase reaction. The light output is proportional to the concentration of succinate from low nM to 15microM. This assay is easy-to-use, fast and simple. The SuccinateGlo JmjC Demethylase Hydroxylase Assay is performed in a single well of a multiwell plate and can be used to detect demethylase dioxygenase activity in as little as a 5microl reaction.
