Recombinant human InsR (amino acids 980-1382-end) was expressed in Sf9 insect cells using a baculovirus and an N-terminal GST tag. InsR is the insulin receptor tyrosine kinase involved in insulin signaling, cleaved post-translationally into alpha and beta chains, which are covalently linked. Insulin binding stimulates glucose uptake, maintaining fuel homeostasis and preventing diabetes. Studies show that conditional knockout of insulin receptor substrate 2 (IRS2) in mouse pancreas beta cells and brain regions, including the hypothalamus, increased appetite, lean and fat body mass, linear growth, and insulin resistance leading to diabetes.
