Recombinant full-length human PKC zeta was expressed by baculovirus in Sf9 insect cells using an N-terminal GST tag. PKC zeta is an atypical isoform of the PKC family. PKC zeta is found in both particulate and soluble fractions and cannot be activated by phorbol ester. Overexpression of PKC zeta and subsequent phorbol ester treatment abolished phorbol ester-induced reduction in cell proliferation. Overexpression of PKC zeta also potentiates phorbol ester-induced mitogen-activated protein (MAP) kinase activation in a PKC-dependent manner. PKC zeta is an upstream modulator of p70S6K, an important regulator of cell proliferation. Kinase Enzyme System contains: Kinase: PKC zeta, 10ug (Human, recombinant full-length). MW: ~93kDa. Substrate: CREBtide (KRREILSRRPSYR); derived from human CREB1 isoform A (amino acid 109-121). Other: Reaction Buffer, DTT. PKC zeta NCBI Database Entry: www.ncbi.nlm.nih.gov/gene/5590/.
