RAF kinases (ARAF, BRAF and CRAF) play an important role in the MAPK signaling pathway, one of the most heavily mutated pathways in cancers. This signaling is typically mediated via the formation of RAF dimers, making these dimers important targets for drug discovery. The NanoBRET TE Intracellular RAF Dimer Assays can be used to quantify compound affinity and fractional occupancy against RAF kinases in live cells. The assays rely on the NanoBRET TE technology, a bioluminescence resonance energy transfer (BRET) method that uses a luciferase as the energy donor and a cell-permeable NanoBRET TE Tracer as the energy acceptor. The NanoBRET TE Intracellular RAF Assay uses the NanoLuc Binary Technology (NanoBiT), instead of NanoLuc-fused target protein as the BRET donor.??
