The Janelia Fluor HaloTag Ligands, including the JFX dyes, are engineered to characterize HaloTag fusion proteins in endogenous settings. The Janelia Fluor HaloTag dyes span the visible spectrum, and their distinct fluorogenic attributes and versatility make them ideal for labeling cell-permeable proteins.The JFX dyes (analogs of the Janelia Fluor dyes) incorporate deuterium into the alkylamino substituents of rhodamines, inhibiting photochemically induced spectral shifts and reducing irreparable photobleaching. The enhanced photostability of these dyes makes them invaluable for detection and single-molecule imaging studies in live cells.A notable feature of these dyes is their ability to support sequential labeling of proteins with different colored ligands via pulse-chase labeling.
