The Bio-Glo-NB VLP Luciferase Assay Systems are optimized for use with HiBiT-PsVLP Bioassays to measure pseudovirus entry into target cells. These assay reagents are functionally tested to ensure consistent performance in detecting luminescence generated by HiBiT and LgBiT complementation. In the presence of neutralizing antibodies or inhibitors, entry or fusion of HiBiT-tagged PsVLPs is blocked, preventing intracellular release of HiBiT and resulting in no luminescent signal. Without inhibition, the PsVLPs bind to target cells and undergo membrane fusion, allowing HiBiT to enter the cells, bind to LgBiT and produce a detectable luminescent signal. This system offers a sensitive and reliable method for evaluating viral entry and inhibition mechanisms in live cells.
